In a new study published today in mBio, a team of researchers from Tel Aviv University and Lund in Sweden have shown that the immune system recognizes and attacks the toxic protein that causes autoimmunity in multiple sclerosis (MS) patients.
"We proved that the immune system recognizes and attacks the toxic protein that causes autoimmunity in multiple sclerosis patients," says Dr. Rafi Yavneh, first author of the study and a research researcher in the Center for Clinical Immunology at TAU's Sackler Faculty of Medicine. "This discovery has been three years in the making and is a significant finding. It helps us understand how the body's own immune system can play an independent role in MS."
MS is a chronic, relapsing, inflammatory disorder of the central nervous system that affects 1 out of every 150 people. During the first relapse, immune system attacks the myelin sheath, protective outer coating of nerve fibers, destroying these cells and leading to permanent disability, or disability progression. There is no cure, and MS symptoms are worsened by certain lifestyle and environmental factors, such as infection, infection-related inflammation and exposure to allergens.
The researchers conducted several studies using mice that were genetically modified to produce high levels of a toxic protein called cetirizin. They found that high levels of cetirizin caused the immune system to react against these mice and destroy myelin. The researchers then treated mice with an antibody that neutralized the cetirizin-induced immune reaction, resulting in the mice regaining health and function.
"The mouse model allowed us to show that the cetirizin-induced immune reaction can lead to the destruction of myelin. Our next step was to determine exactly what triggers this reaction," says first author Dr. Elisabeth Schmiedt-Pfeifer, TAU research scholar in the Sackler Faculty of Medicine. "We found that myelin destruction is triggered by cetirizin, but not other proteins that are important components of the immune system."
The researchers also found that protein is responsible for the immune reaction is not found in the plasma of normal people.
